Janet Lupo was born and raised in Hoboken, New Jersey, United States. Her parents were Giuseppe and Pauline Lupo.
She attended Hoboken High School. After graduating from Hoboken High School, Janet majored in cosmetology at the Parisian Beauty Academy in Hackensack, New Jersey.
In April 1974, Lupo secured a position as a Playboy Bunny at the resort’s Great Gorge Hotel in McAfee, New Jersey.

While working there, she started posing for Playboy’s Playmate of the month. In November 1975, Janet Lupo was featured as the Playmate of the Month.
Janet Lupo traveled and promoted Playboy in Canada, Japan, and the US after her Playmate tenure. She relocated to Key Biscayne, Florida, in January of 1977 and started working as a flight attendant there.
In the early 1980s, Lupo appeared in multiple Playboy reunion photo shoots. Later, Lupo made appearances at the Glamourcon conventions in Chicago and Los Angeles.
